Gwen Stefani terlahir di sebuah keluarga campuran dari orang tuanya yang berdarah Italia (ayahnya) dan
Britania Raya (ibunya). Ia memiliki seorang adik perempuan Jill dan dua saudara lelaki bernama Eric dan Todd.
Kakak lelakinya Eric dan kawannya John Spence pada tahun 1987 memulai band No Doubt.
Namun tidak lama kemudian John Spence bunuh diri dan Gwen menjadi vokalis satu-satunya. Kemudian selang beberapa lama kemudian kakaknya, Eric Stefani yang memainkan kibor pergi untuk menjadi penggambar serial The Simpsons pada tahun 1994 . Eric juga kurang puas kala itu dengan perkembangan band mereka.
Pada tahun 2004 Gwen mencoba sebuah karier solo dan ternyata sukses.
